13.5 billion U.S. dollars will be invested in the project.
Government officials accepted the proposal after a panel outlined measures to combat gamling addiction and the financial stability of the businesses involved.
Prime Minister Fumio Kishida says he hopes the resort will contribute to the development of the region and Japan as a whole after Osaka holds the World Expo in 2025.
It is also expected to attract domestic and international tourist spending.
Casinos and private gambling had been illegal in Japan until a 2018 integrated resort law provided exception to casino games like poker at officially approved establishments aimed at attracting tourists to the country.
