Israeli officials say the two bearded gunmen, who were then killed by two police officers nearby, were supporters of the Islamic State extremist group.
They were Arab citizens from northern Israel.
While there's still no claim of responsibility, Hamas, the Islamic militant group that rules the Gaza Strip, praised the attack as a "heroic operation".
A man and a woman were killed, with four others wounded, two of whom seriously.
Israeli Foreign Affairs Minister Yair Lapid condemned the attack as heinous and savage committed aimed at terrorising and damaging the fabric of Israel's society.
The attack, which occurred prior to the volatile period of Ramadan, is expected to overshadow a meeting of foreign ministers in the Negev desert, in which the Iranian nuclear deal is due to be discussed.
